The Detroit Lions and Buffalo Bills meet on Thursday Night Football, with kickoff set for 8:15 p.m. ET at the new Highmark Stadium on September 17. Buffalo enters Week 2 after an impressive road win over the Texans, while Detroit needed overtime to escape with a 31-30 win over the Saints.
With both teams considered Super Bowl contenders, could this be an early preview of Super Bowl LXI? In this article, we’ll break down the biggest storylines and explain why our model likes Buffalo to cover 4.5 points.
ATS Expert Verdict:

“I like the Bills -4.5 here. Detroit has been by far the NFL’s best team ATS since Dan Campbell took over in 2021, but I expect some regression this year and beyond. Their defense still looks vulnerable, and I’m comfortable laying the points with Buffalo on a short week in what should be an emotional home opener at their new stadium.”
Polymarket Pricing
Polymarket currently has Buffalo priced around 49¢ to cover the 4.5-point handicap, while our model gives the Bills a 56% chance to cover. That gives us a 7% edge at the current price, with the model projecting a 29-24 Buffalo win.
Josh Allen Should Find Success Through the Air
Detroit struggled against the pass last season, and those problems showed up again in Week 1. Second-year quarterback Tyler Shough threw for 410 yards and three touchdowns, which could spell trouble against Josh Allen.
Can Detroit Exploit Buffalo’s Run Defense?
Buffalo’s run defense remains a weakness, but Detroit may have a hard time taking advantage on a short week. Jahmyr Gibbs just had a huge workload with 29 carries and five catches on Sunday, while expected backup Isiah Pacheco is currently on IR.

One of the biggest questions coming into this matchup is whether Detroit’s secondary can hold up against Josh Allen. The 2024 MVP opened the season with 334 yards and two touchdowns in Buffalo’s 36-31 win over Houston, averaging 11.5 yards per attempt.
That could be a problem for a Lions defense that allowed the third-most passing yards in the NFL in 2025 (4,567). Detroit struggled again in Week 1, allowing Tyler Shough to throw for 410 yards and three touchdowns.
The Lions nearly blew a 21-0 lead and needed a stop on a two-point conversion in overtime to hold onto the win. If those problems continue, Allen should have plenty of opportunities to attack Detroit through the air.
Thursday night will be a historic one in Buffalo, as the Bills play their first regular-season game at the new Highmark Stadium. The $2.1 billion stadium was built right across the street from their old home, where the Bills played for more than 50 years.
There probably couldn’t be a much bigger stage for the opener, either. Buffalo gets a standalone primetime game against another Super Bowl contender, with Bills Mafia getting its first chance to pack the 60,108-seat stadium for a game that counts.
It’s tough to put a number on how much that matters, but I expect an emotional atmosphere and a Buffalo team that will be focused on giving its fans a win. On a short week against a Detroit team coming off an overtime game, that could give the Bills another small edge.
Detroit is already dealing with several key injuries heading into Week 2. Starting safeties Brian Branch and Kerby Joseph remain on the PUP list, leaving an already vulnerable secondary without two of its best players against Josh Allen and the Bills.
The Lions are also banged up along the offensive line. Starting center Cade Mays has a broken wrist, while guard Christian Mahogany left Sunday’s win and didn’t return. His status will be worth watching on a short week.
Buffalo is in better shape overall, though it has a few injuries to watch. Ty Johnson missed Week 1 with a hamstring injury, while defensive tackle T.J. Sanders was inactive with a knee injury. D.J. Moore also went to the medical tent with a lower-leg injury late against Houston, but appears to have avoided any significant injury.
There’s a lot to like about Buffalo in this spot. Josh Allen is 3-0 in his career against Detroit, and the Lions’ injuries and problems in the secondary give him a good chance to keep that streak going.
The 4.5-point handicap doesn’t scare me much, either. Seven of Detroit’s eight losses last season came by at least five points, and I expect Buffalo to create enough separation at home to win by 5+.
Could we still see these teams meet again in Super Bowl LXI? Absolutely, but Detroit will need to get healthier and show some improvement defensively if it wants to make another deep run. For Thursday night, I’m giving the edge to Buffalo.
Prediction: Bills 29, Lions 24
Best Trade: Buffalo Bills -4.5 at 49¢ (playable up to 55¢)
